Description

Genalyte’s connective tissue disease kits measure autoantibodies reactive with SS-A 60, SS-A 52, SS-B, Sm, RNP, Scl-70, Jo-1, dsDNA, ribosome-P, and centromere A+B.  Antibodies to these anti-nuclear antigens have been associated with a range of connective tissue diseases including systemic lupus erythematosus (SLE), Sjögren’s syndrome, mixed connective tissue disease, systemic sclerosis, polymyositis, and dermatomyositis. These kits can be used for research into these and other connective tissue diseases using a small sample volume (<5 uL) and can provide results within 15 minutes.

Principle of the Method

Genalyte chips contain a series of silicon biosensors on the chip surface, which are functionalized with autoantigens.  A sample such as serum, plasma, or whole blood is flowed over the antigens. If an autoantibody specific for the antigen is present, it will bind.  The specificity of binding is confirmed and the signal corresponding to the bound antibody can be amplified with the use of secondary detection reagents for total Ig or specific isotypes.

Background Information

Autoimmunity is the failure of an organism’s immune system to recognize its own constituent parts as “self,” allowing an immune response against its own cells and tissues. Prominent examples of autoimmune diseases include systemic lupus erythematosus (SLE), Type 1 Diabetes, Rheumatoid Arthritis and Sjögren’s Syndrome.
